InSillyClo is a time-saving tool for efficient cloning workflows at any scale. It provides a complete environment for in silico cloning and related calculations, allowing users to focus on their designs rather than repeating each time manually side tasks.
Key features:
- Assembly designer (optional): This module lets users define the assembly type by managing the number of input parts, their role, and the naming convention. Defining tailored assemblies increases clarity, thereby avoiding mistakes. After assembly definition, users can download template file to be filled at each cloning workflow with plasmids to build.
- Assembly simulator (core): At the heart of InSillyClo, this module performs the in silico assembly. In this module, users upload the DB_oP file with plasmids to build, along with input plasmid data, their sequences and the mapping between names and identifiers. Once the assembly of the output plasmid performed, users can simulate colony PCR and restriction digestion, compute dilutions and master mixes, and download the SBOL file of the assembly.
Extra tools include agarose gel simulations (post-PCR or restriction digest) and input plasmid dilution calculations for equimolar mixing. Assembly Simulator is versatile, supporting both small and large projects, and can run independently of Assembly Designer, ideal for beginners. learn more
